Ce bâtiment a été habité par trois générations de la famille Colón. Sa construction a commencé à la fin de 1510 et en 1512, Don Diego Colón, vice-roi des Indes, troisième gouverneur d'Hispaniola et fils du découvreur de l'Amérique, y vivait déjà.
L'Alcázar est un bâtiment de forme rectangulaire à deux niveaux et a été construit en pierre coraline provenant des carrières de Santa Bárbara.
Actuellement, c'est un musée lié à la période vice-royale en République dominicaine.
