Il palazzo fu edificato dalla famiglia Lanfranchi all'inizio del XVI secolo e assunse l'aspetto attuale alla fine del secolo dopo i restauri: intonaci in frassino, edicole e ginocchiere. Il nome Tuscanylli fu coniato nel 1827 quando il palazzo fu acquistato dall'omonima famiglia. All'interno i soffitti presentano affreschi di Nicola Cianfanelli, Gaspero Martellini e Annibale Gatti raffiguranti scene di Byron e poesie, L'Apoteosi di Galileo e L'Apoteosi di Michelangelo (al quale si dice sia stato attribuito il progetto dell'edificio).
