I 1011 blev byen første gang nævnt i litteraturen, da bygningen med navnet Novum Castellum blev givet af den burgundiske konge Rudolf III til sin kone.
I 2002 var Neuchâtel en af værtsbyerne for Expo.02. Byen er også kendt for sin urindustri, og fordi en type ostefondue er opkaldt efter byen. Desuden blev Marlboro-cigaretter her produceret fra 1924.
