Die zweite Etappe ist 62 km lang. Nachdem man von Setúbal mit der Fähre übergesetzt ist, führt die Route von Tróia nach Santiago do Cacem. Der erste Teil verläuft durch eine dünenartige, flache Landschaft, später wird es hügeliger, und in den letzten 5 km gibt es einen ernsthaften Anstieg nach Santiago.
