Tijdens de Renaissance (15e-16e eeuw) werd Florence geregeerd door de familie De' Medici. Onder hun bescherming bloeiden de kunsten, wetenschap en literatuur als nergens anders in Europa. Florence was de stad van schrijvers als Dante, Petrarca en Macchiavelli, en van kunstenaars en ingenieurs zoals Boticelli, Brunelleschi (die de prachtige koepel van de kerk Santa Maria della Fiori bouwde), Alberti, Leonardo da Vinci en Michelangelo. Ook Galileo kon rekenen op de steun van deze familie. Hij schonk ook veel van zijn gereedschap aan de Medici. Een deel ervan is nog steeds te zien in het Museo della Scienza, aan de rivier de Arno. De Medici vroegen Brunelleschi om de kerk van San Lorenzo te bouwen.
