Partenstein er en kommune i det underfrankiske distrikt Main-Spessart. Den navngivne sogneby er sæde for kommunalbestyrelsen og forvaltningssamfundet Partenstein. Den føderale vej 276 går gennem byen.
Partenstein ligger i regionen Main Spessart midt i Spessart ved den venstre bred af Lohr, cirka syv kilometer nordvest for byen Lohr am Main. Det topografisk højeste punkt i kommunens område ligger med 502 m over NHN på Gaulskopf, nordøst for Partenstein, det laveste ligger ved Lohr på 174 m over NHN. Kahltal-Spessart-cykelruten går gennem byen.
