Klosteret Santo Domingo Real, der ligger i byen Toledo i Castilien-La Mancha, Spanien, er et kloster grundlagt i 1364 af adelskvinden Inés García de Meneses, datter af García Suárez de Meneses og María Fernández Barroso, efter at hun blev enke efter Sancho de Velasco.
Det har været et lukket kloster siden dets begyndelse og det første i byen, grundlagt af de dominikanske nonner. Her professerede María de Castilla. Maria var datter af kong Peter af Castilien og hans elskerinde Teresa de Ayala. Teresa var datter af Diego Gómez de Toledo og hans kone Inés de Ayala, kansler Pero López de Ayala's søster.
