Byen Castiglion Fiorentino ligger på en bakke 342 m over havets overflade, 17 km sydøst for Arezzo. Afgrænset mod øst af Preappennini, strækker det kommunale område sig delvist over Valdichiana og de højer, der ligger ud for, og delvist omfatter næsten hele bassinet i Val di Chio, inklusive den øverste del af dalen af floden Nestore, mens en lille sektion strækker sig over bunden af torrenten San Chimento, der løber ud i Cerfone og derefter til Tevere, på den anden side af Foce-passet. Området grænser til kommunerne Arezzo mod nord, Cortona mod øst og syd, Foiano della Chiana mod sydvest og Marciano della Chiana mod vest.
