La rivière Eselsbach prend sa source dans la forêt palatine du nord-ouest, à la limite ouest du village d'Enkenbach-Alsenborn, à une altitude de 304 m au-dessus du niveau de la mer. De là, le ruisseau s'écoule principalement vers l'ouest et se jette dans la Lauter sur la droite, au nord-ouest de Kaiserslautern, à 226 m.
Après un trajet de 12,3 km, l'Eselsbach se jette 78 mètres plus bas, ce qui correspond à une pente moyenne de lit de 6,3 ‰. Il draine un bassin versant de 27,888 km² via la Lauter, le Glan, la Nahe et le Rhin vers la mer du Nord.
