Le palais a été construit par la famille Lanfranchi au début du XVIe siècle et a acquis son aspect actuel à la fin du siècle après des rénovations : enduits de frêne, édicules et fenêtres à pans coupés. Le nom de Tuscanylli a été inventé en 1827 lorsque le palais a été acheté par la famille du même nom. À l'intérieur, les plafonds présentent des fresques de Nicola Cianfanelli, Gaspero Martellini et Annibale Gatti représentant des scènes représentant Byron et la poésie, L'Apothéose de Galilée et L'Apothéose de Michel-Ange (à qui la conception du bâtiment aurait été attribuée).
