De Ezelbeek ontspringt in het noordwestelijke Pfälzerwald aan de westelijke rand van Enkenbach-Alsenborn op een hoogte van 304 m boven NHN. Van hieruit stroomt de beek voornamelijk in westelijke richtingen en mondt in het noordwesten van Kaiserslautern op 226 m rechts in de Lauter.
Na zijn 12,3 km lange weg mondt de Ezelbeek 78 hoogtemeters dieper uit, wat overeenkomt met een gemiddelde helling van 6,3 ‰. Hij voert een oppervlakte van 27,888 km² af via Lauter, Glan, Nahe en Rijn naar de Noordzee.
