Die erste Besiedlung des heutigen Stadtgebiets fand Anfang des 17. Jahrhunderts statt. 1705 wurde Brookline zur Stadt erklärt und hat es seither verstanden, seine Selbständigkeit gegenüber der Großstadt Boston zu behaupten, die Brookline fast vollständig umschließt. Brookline ist der Geburtsort der Brüder John und Robert F. Kennedy.
