Die Klosterstraße ist eine der ältesten Berliner Straßen im Ortsteil Mitte. Der heutige Name geht auf das Graue Kloster, ein ehemaliges Franziskanerkloster, in der Straße zurück, von dem heute nur die Ruine der Klosterkirche erhalten ist. Nach der Straße ist wiederum das Klosterviertel benannt, in dem sich zahlreiche Baudenkmäler befanden, die zum Teil noch erhalten sind.
Die Klosterstraße ist eine ruhige und etwa 430 Meter lange Querstraße der Grunerstraße. Sie verläuft in einem leichten Bogen nach Süden bis zur Spree, kreuzt dabei die Parochialstraße und die Stralauer Straße und geht bis zum Rolandufer.
