Die Gemeinde Guillestre liegt auf einer durchschnittlichen Höhe von 1.000 m und überragt das linke Ufer der Schluchten des Guil, kurz bevor sie mit der Durance zusammenfließt. Sie liegt am Tor zum Queyras und ist der einzige Zugang zu diesem Tal, insbesondere in den Wintermonaten (der Col Agnel und der Col Izoard sind einen Teil des Jahres geschlossen).
Guillestre ist ein Knotenpunkt und Ausgangspunkt für den Col d'Izoard, den Col de Vars und den Col Agnel. Sie ist auch ein Durchgang zwischen Embrun und Briançon.
