El municipio de Guillestre se encuentra a una altitud media de 1.000 m y domina la orilla izquierda de los desfiladeros del Guil, justo antes de unirse al Durance. Situado en la puerta de la Queyras, es el único punto de acceso a este valle, especialmente durante los meses de invierno (el col Agnel y el col Izoard están cerrados parte del año).
Guillestre es un cruce y un punto de partida para el col d'Izoard, el col de Vars y el col Agnel. También es un paso entre Embrun y Briançon.
