Der Bahnhof Comber lag an der Belfast und County Down Railway, die von Belfast nach Newcastle, County Down in Nordirland führte.
Der Bahnhof wurde am 6. Mai 1850 von der Belfast und County Down Railway als vorletzter Bahnhof der ursprünglichen eingleisigen Linie nach Newtownards eröffnet.
Mit der Eröffnung der Zweigstrecke nach Ballygowan und Downpatrick im Jahr 1958 wurde er zu einem Knotenpunkt. Der Zweig nach Newtownards wurde schließlich bis Donaghadee und die Linie nach Downpatrick bis Newcastle verlängert.
