You can start this route anywhere. The castle (Schauenburg) is definitely a good starting point (and place to relax). Along this bike route lies a religious site: Evangelical Church. A perfect spot to rest. You can start this route at the museum (Grube Anna-Elisabeth), but that is not necessary. Almost forgot, make sure to also stop by the Jewish Cemetery Schriesheim.
