Les Jésuites s'installent d'abord dans les bâtiments de l'ancien collège du Quercy (XVIe siècle). Ils modifient l'ancienne chapelle Saint-Michel-des-Pauvres, qu'ils consacrent désormais à Ignace de Loyola, fondateur de l'ordre, et confient la réalisation de son décor peint à deux Jésuites : François Blanc et Jean Planchette.
Les Jésuites font bâtir deux imposants corps de bâtiments, à la jonction desquels s'implante un grand escalier à volées droites. Au centre de l'établissement, la salle de Déclamation possède un remarquable plafond à la française.
En 1676, la construction du campanile à la toulousaine dominant le collège est confiée à l'architecte Antoine Chassagnard.
