De Posthoorn behoorde oorspronkelijk toe aan schout Willem de Clerck, die in het gebouw zijn hof gevestigd had.
Vanaf 1588 vond het klooster van Bethaniën hier dan weer onderdak, tot de kloosterorde in 1783 onder Frans bewind opgeheven werd en het pand in particuliere handen terechtkwam. Zoals we zo vaak zien heeft de kloostergemeenschap het klooster opnieuw opgekocht.
Een deel van de gebouwen werd door J.F. Scheppers verworven die het verbouwde tot brouwerij "De Posthoorn".
