Dit gebouw werd bewoond door drie generaties van de familie Colón. De bouw begon aan het einde van 1510 en in 1512 woonde daar Don Diego Colón, vicekoning van de Indiën, de derde gouverneur van Hispaniola en de zoon van de ontdekker van Amerika.
Het Alcázar heeft een rechthoekige plattegrond met twee niveaus en is gebouwd met koraalsteen afkomstig uit de steengroeves van Santa Bárbara.
Tegenwoordig is het een museum dat verband houdt met de vicekoninklijke periode in de Dominicaanse Republiek.
