De gemeente Guillestre ligt op een gemiddelde hoogte van 1.000 m en domineert de linkeroever van de kloven van de Guil, kort voordat deze samenkomt met de Durance. Gelegen aan de poort van de Queyras, is het het enige toegangspunt tot deze vallei, vooral tijdens de wintermaanden (de col Agnel en de col Izoard zijn een deel van het jaar gesloten).
Guillestre is een kruispunt en een vertrekpunt voor de col d'Izoard, de col de Vars en de col Agnel. Het is ook een doorgang tussen Embrun en Briançon.
