De poort is gebouwd rond 1350 als onderdeel van een stenen vestingmuur met omgrachting. Door uitbreiding van de vesting in 1484 komt de poort binnen de muren van de stad te liggen. Daardoor verliest ze haar functie als toegangspoort en verdedigingswerk. Ondertussen is de poort al enige tijd een gevangenis, waardoor ze bespaard blijft van de sloop. Tot ongeveer 1925 wordt de poort nog als gevangenis gebruikt.
In 2015 is de Gevangenpoort gerestaureerd en heringericht waardoor de verhalen van de poort en haar bewoners nog beter beleefd kunnen worden. Sinds juli 2015 is de Gevangenpoort ook een buitenpost van stadspaleis en museum het Markiezenhof.
