De 1575 à 1622, Aigues-Mortes était l'un des huit refuges sûrs accordés aux Protestants. La révocation de l'Édit de Nantes en 1685 a entraîné une sévère répression du protestantisme, marquée en Languedoc et dans les Cévennes au début du XVIIIe siècle par la "guerre des Camisards". Comme d'autres tours de la ville, à partir de 1686, la tour Constance a été utilisée comme prison pour les Huguenots qui refusaient de se convertir au catholicisme romain. En 1703, Abraham Mazel, leader des Camisards, a réussi à s'échapper avec seize compagnons.
