C'est ainsi le plus petit quartier du district de Rotterdam-Nord. Le quartier est délimité par la Schiekade, la Bergweg, la Noordsingel et la Heer Bokelweg.
Là où se trouve maintenant le quartier Agniesebuurt, se trouvait auparavant la poldre Oost-Blommersdijk. Dans cette zone entre la Rotte et la Schie de Rotterdam, il y avait déjà quelques maisons au XVIIIe siècle, mais entre 1850 et 1940, toute la poldre a été construite. Le quartier Agniesebuurt est nommé d'après la rue Agniesestraat, située au milieu. Le nom Agniese provient d'Agniese, la seule fille de Dieric Bokel. Elle épousa en 1336 Simon van Benthem, puis Gherijt van Herlaer. Par ses mariages, la plus grande partie des biens de son père est passée à d'autres familles.
