Het klooster van Santo Domingo Real, gelegen in de stad Toledo, in Castilië-La Mancha, Spanje, is een convent dat in 1364 is opgericht door de nobele vrouw Inés García de Meneses, dochter van García Suárez de Meneses en María Fernández Barroso, nadat ze weduwe was geworden van Sancho de Velasco.
Het is sinds zijn ontstaan een gesloten klooster en het eerste in de stad dat is opgericht door de Dominicaanse nonnen. Hier heeft María de Castilla haar geloften afgelegd. Maria was de dochter van koning Peter van Castilië en zijn minnaar Teresa van Ayala. Teresa was de dochter van Diego Gómez de Toledo en zijn vrouw Inés de Ayala, de zuster van kanselier Pero López de Ayala.
