Spoleto è situata all'estremità meridionale della Valle Umbra, vasta pianura alluvionale, generata in epoca preistorica dalla presenza di un vasto lago, il lacus Umber, prosciugato definitivamente nel Medioevo, dopo il suo impaludamento, con delle opere di bonifica.
La città si è sviluppata sul colle Sant'Elia, un basso promontorio collinare alle falde del Monteluco, nei pressi del fiume Clitunno, e più in basso fino alle rive del torrente Tessino; ad est è contornata dai monti che delimitano la Valnerina, di cui tre delle sue frazioni, Le Cese, Belvedere e Ancaiano, ne fanno parte.
