Moulsford station var på den oprindelige rute af Great Western Railway og var en af de tre mellemstationer, der blev oprettet, da linjen blev forlænget fra Reading til Steventon i 1840.
Great Western Railway blev bygget og åbnet i etaper. Det blev åbnet så langt som til Reading den 30. marts 1840; den 1. juni 1840 blev det åbnet til Steventon med tre mellemstationer, hvor den nordligste var Wallingford Road; det blev muligvis oprindeligt navngivet Moulsford, og blev omdøbt i december 1840.
