Il comune di Guillestre si trova a un'altitudine media di 1.000 m e domina la riva sinistra delle gole del Guil, poco prima che si unisca al Durance. Situato alla porta del Queyras, è l'unico punto di accesso a questa valle, soprattutto durante i mesi invernali (il colle Agnel e il colle Izoard sono chiusi per parte dell'anno).
Guillestre è un incrocio e un punto di partenza per il col d'Izoard, il col de Vars e il col Agnel. È anche un passaggio tra Embrun e Briançon.
