Paladset blev bestilt af Domenico della Rovere, kardinalpræst af San Clemente og nevø af pave Sixtus IV., og blev mellem 1480 og 1490 udført af arkitekten Baccio Pontelli i Borgo. Den florentinske arkitekt Pontelli orienterede sig tæt efter den arkitektoniske stil fra Palazzo Venezia, en af de vigtigste bygninger i det 15. århundrede i Rom. I over 300 år var paladset kendt som Palazzo dei Penitenzieri. Paladset er fredet og kan kun besigtiges i hotelområdet.
