Teatro Caio Melisso er et operahus beliggende i Spoleto, Italien, og det fungerer som hovedsted for operaopførelser under den årlige sommerfestival Festival dei Due Mondi.
Teatret har gennemgået flere forvandlinger og navneændringer siden slutningen af det 17. århundrede. Teatro di Piazza del Duomo, også kendt som Teatro della Rosa, bygget omkring 1667, blev moderniseret i 1749 og genåbnet i 1749 som Nuovo Teatro di Spoleto. Efter en restaurering i 1817 og efter at Rossini havde besøgt teatret, faldt det gradvist i unåde, og et nyt teater var nødvendigt i midten af det 19. århundrede. Det 800-sæders Teatro Nuovo blev bygget mellem 1854 og 1864 for betydelige offentlige udgifter.
