Det blev grundlagt i det 15. århundrede af familien Heringa. I 1603 kom det i besiddelse af familien Van Eysinga, og i 1631 blev det grundigt ombygget. I 1687 blev det købt af Dr. Henricus Popta. Efter hans død i 1712 blev Poptaslot testamenteret til Poptagasthuis, et hvilehjem for fattige kvinder. Fra 1906 til 1908 blev Poptaslot restaureret og ombygget efter et design af den Haag-arkitekt Johan Frederik Lodewijk Frowein. Huset er stadig møbleret og indrettet i 17. og 18. århundredes stil.
