Die Kapelle ist der einzige verbliebene Teil des 1505 gegründeten Colegio de Santa María de Jesús.
Das Kapellengebäude ist im spätgotischen Mudéjar-Stil gehalten und weist Teile aus Sichtziegel auf.
Der Bau begann im Jahr 1506. Der Entwurf wird Alonso Rodríguez zugeschrieben. Im Jahr 1509 leitete Antón Ruiz die Arbeiten. Die Fassade wurde 1514 vom Maurer Manuel Sánchez angefertigt.
Das Colegio de Santa María de Jesús begann im Jahr 1516, Studenten aufzunehmen.
Das Hauptaltarbild wurde 1520 von Alejo Fernández geschaffen. Es zeigt die Virgen de la Antigua, die ein Modell des Kollegs erhält, das ihr von dessen Gründerin, Maese Rodrigo Fernández de Santaella, geschenkt wurde.
Bemerkenswert sind auch die Kachelszenen in „Cuerda Seca“ und die Polychromie.
