La chapelle est la seule partie restante du Colegio de Santa María de Jesús, fondé en 1505.
Le bâtiment de la chapelle est de style gothique tardif-mudéjar , avec des parties en briques apparentes.
Sa construction fut commencée en 1506. La conception est attribuée à Alonso Rodríguez. En 1509, Antón Ruiz fut chargé des travaux. La façade a été réalisée en 1514 par le maçon Manuel Sánchez.
Le Colegio de Santa María de Jesús commença à recevoir des étudiants en 1516.
Le retable principal a été réalisé en 1520 par Alejo Fernández. Il représente la Virgen de la Antigua recevant une maquette du collège que lui a présentée son fondateur, Maese Rodrigo Fernández de Santaella.
Les scènes de carreaux, en cuerda seca et polychromes, sont également remarquables.
