De kapel is het enige overgebleven deel van het Colegio de Santa María de Jesús, gesticht in 1505.
Het kapelgebouw is laatgotisch-Mudejarstijl, met delen in zichtbare baksteen.
Er werd mee begonnen in 1506. Het ontwerp wordt toegeschreven aan Alonso Rodríguez. In 1509 had Antón Ruiz de leiding over de werkzaamheden. De gevel werd in 1514 gemaakt door de metselaar Manuel Sánchez.
Het Colegio de Santa María de Jesús begon studenten te ontvangen in 1516.
Het hoofdaltaarstuk werd in 1520 gemaakt door Alejo Fernández. Het stelt de Virgen de la Antigua voor die een model van het college ontvangt dat haar werd aangeboden door de stichter, Maese Rodrigo Fernández de Santaella.
Ook opmerkelijk zijn de tegeltableaus, in cuerda seca en polychroom.
