Kapellet er den eneste tilbageværende del af Colegio de Santa María de Jesús, grundlagt i 1505.
Kappelebygningen er sen-gotisk-Mudejar stil, med dele i synlig mursten.
Det blev påbegyndt i 1506. Designet tilskrives Alonso Rodríguez. I 1509 havde Antón Ruiz ansvaret for arbejderne. Facaden blev i 1514 lavet af mureren Manuel Sánchez.
Colegio de Santa María de Jesús begyndte at tage imod studerende i 1516.
Hovedalteret blev lavet i 1520 af Alejo Fernández. Det forestiller Virgen de la Antigua, der modtager en model af kollegiet, som blev tilbudt hende af grundlæggeren, Maese Rodrigo Fernández de Santaella.
