Oudesluis emerged fairly shortly after the construction of the Zijpe polder at the Groote Sluys built there. The village name is likely derived from the fact that the sluice was one of the first sluices in the area. The sluice was initially made of wood but was replaced in 1631 by a larger stone one and was then one of the largest sluices in Europe.
At that time, Oudesluis was located at the Wadden-like outflows of the former Zuiderzee and was a docking place for VOC ships and also the home port for a fishing fleet. When the Anna Paulownapolder was constructed in 1847, Oudesluis became cut off from the sea.
