Byen ligger på den nordøstlige kant af Harzen. Selke løber gennem stedet, hvor flere damme ligger. Der hører også Pappelteich til.
Ermsleben blev i år 1045 under navnet "Anegremislebo" første gang nævnt som ejet af grevskabet Falkenstein. Allerede i 1298 blev stedet betegnet som "oppidum". I 1296 nævnes den sandsynligvis ældre kirke St. Sixtus, som senere blev ombygget flere gange, for første gang. Stedet fik markedretten i 1530.
