Situato nel cuore della pianura di Brienne e sul percorso della strada romana Troyes-Montier en der, Perthes les Brienne non è sempre stato conosciuto con questo nome. I documenti del 1632 riportano il villaggio come Perthes en Rothière fino al 1919. Nel Medioevo, la signoria di Perthes apparteneva all'Abbazia di Boulancourt nel comune di Longeville sur la Laines, nella Haute-Marne. Oggi Perthes è rinomata per la produzione di cavoli per crauti. Del passato rimane la chiesa a graticcio di Saint Denis, risalente al XVI secolo. Sono visibili anche frammenti di vetrate del XVI secolo.
