Het station werd in 1837 door de Staatsspoorwegen gebouwd. Kort na de onaf-hankelijkheid besliste België onder impuls van koning Leopold I om als eerste staat spoorlijnen aan te leggen. Melle lag langs de derde aangelegde hoofdlijn: Mechelen-Gent (na Brussel en Antwerpen), waardoor het ook het oudste spoorstation van België en bij uitbreiding dus ook het oudste bakstenen station van de wereld had. De meeste stations werden echter toen in hout gebouwd omdat dit goedkoper was.
