La gare a été construite en 1837 par les Chemins de fer d'État. Peu après l'indépendance, la Belgique, sous l'impulsion du roi Léopold I, a décidé d'installer des voies ferrées en tant que premier État. Melle se trouvait sur la troisième ligne principale construite : Malines-Gand (après Bruxelles et Anvers), ce qui lui a également permis d'avoir la plus ancienne gare de Belgique et, par extension, la plus ancienne gare en briques du monde. Cependant, la plupart des gares étaient alors construites en bois car c'était moins cher.
