Karlshof er en del af den amtsangehörige kommune Neulewin i Landkreis Märkisch-Oderland. Kommunen Neulewin administreres af Amt Barnim-Oderbruch.
Karlshof lå i det 18. århundrede på området af landsbyen Güstebiese. Det tilhørte Johanniterkomturei Grüneberg. I 1756 blev forarbejdet Carlshof grundlagt. I 1799 tilhørte der godset ti boligkasser til familier. I 1837 blev der indrettet et bønsal i skolen. Før det blev der i 1838 opsat en klokkestol, som blev nedtaget i 1972 på grund af bygningsmæssige problemer. I 2003 blev der opført en ny klokkestol til klokken fra 1837.
