La capilla es el único resto que queda del Colegio de Santa María de Jesús, fundado en 1505.
El edificio de la capilla es de estilo gótico-mudéjar tardío , con partes en ladrillo visto.
Se inició en 1506. El diseño se atribuye a Alonso Rodríguez. En 1509 Antón Ruiz se hizo cargo de la obra. La fachada fue realizada en 1514 por el albañil Manuel Sánchez.
El Colegio de Santa María de Jesús comenzó a recibir alumnos en 1516.
El retablo mayor fue realizado en 1520 por Alejo Fernández. Representa a la Virgen de la Antigua recibiendo una maqueta del colegio que le regaló su fundador, Maese Rodrigo Fernández de Santaella.
Destacan también las escenas de azulejos, de cuerda seca y policromadas.
