La capella és l'única part que queda del Col·legi de Santa María de Jesús, fundat l'any 1505.
L'edifici de la capella és d'estil gòtic-mudèjar tardà , amb parts en maó vist.
Es va iniciar l'any 1506. El disseny s'atribueix a Alonso Rodríguez. L'any 1509 Antón Ruiz s'encarregava de l'obra. La façana va ser realitzada l'any 1514 pel paleta Manuel Sánchez.
El Colegio de Santa María de Jesús va començar a rebre estudiants l'any 1516.
El retaule major va ser realitzat l'any 1520 per Alejo Fernández. Representa la Mare de Déu de l'Antigua rebent una maqueta del col·legi que li va regalar el seu fundador, Maese Rodrigo Fernández de Santaella.
També destaquen les escenes de rajoles, en cuerda seca i policromades.
